چکیده

SEQUENCE ORIGIN: Small RNA molecule U6 was isolated from the total cellular RNA of vegetative Phvsarnm microplasmodia (1) by preparative polyacrylamide electroshoresis (2,3). The molecule was radiolabeled in vitro to high-specific-activity using [ T>)-pCp and RNA-ligase, and sequenced by direct chemical methods as we previously described (3). In this sequencing process, pseudouridine scores as a uridine, methylcytosine scores as a cytosine, and methyladenosine scores as an adenosine.

Primary and hypothetical secondary structure of Drosophila melanogaster U6 RNA.

A comparison of the nucleotide sequence of Drosophila U6 RNA with that of rat (6) reveals a homology of 90.7%. Of the 10 differences, 4 represent sugar and base modifications (positions 64, 69, 71, 73) while 6 are nucleotide replacements (positions 4, 6,18,65,86,102). Multiple alignements show that the nucleotide sequences of broad bean (7) and Physarum polycephalum U6 snRNA (8) have much weake...

Proximity of conserved U6 and U2 snRNA elements to the 5' splice site region in activated spliceosomes.

Major structural changes occur in the spliceosome during its catalytic activation, which immediately precedes the splicing of pre-mRNA. Whereas changes in snRNA conformation are well documented at the level of secondary RNA-RNA interactions, little is known about the tertiary structure of this RNA-RNA network, which comprises the spliceosome's catalytic core. Here, we have used the hydroxyl-rad...

A U6 snRNA gene with an internal promoter is juxtaposed to an snRNP protein sequence within an intron of a human G protein gene.

A complex locus on human chromosome 1 brings together sequences homologous to a G protein and two components of the RNA processing machinery of eukaryotic cells. Specifically, the seventh intron of the human Gi3 alpha gene contains a fusion of a partial snRNP E protein pseudogene to a variant U6 snRNA gene. The novel U6 sequence contains nine point mutations and a one nucleotide deletion relati...

The 5' and 3' domains of yeast U6 snRNA: Lsm proteins facilitate binding of Prp24 protein to the U6 telestem region.

The 5' and 3' domains of yeast U6 snRNA contain sequences that are thought to be important for binding to Prp24 and Lsm proteins. By extensive mutational analysis of yeast U6 snRNA, we confirmed that the 3' terminal uridine tract of U6 snRNA is important for U6 binding to Lsm proteins in yeast. Binding of Prp24 protein to U6 RNA is dependent on or is strongly enhanced by U6 binding of Lsm prote...

PRP38 encodes a yeast protein required for pre-mRNA splicing and maintenance of stable U6 small nuclear RNA levels.

An essential pre-mRNA splicing factor, the product of the PRP38 gene, has been genetically identified in a screen of temperature-sensitive mutants of Saccharomyces cerevisiae. Shifting temperature-sensitive prp38 cultures from 23 to 37 degrees C prevents the first cleavage-ligation event in the excision of introns from mRNA precursors. In vitro splicing inactivation and complementation studies ...
